Er befasste er sich insbesondere mit der Physiologie des Blutkreislaufs, Ernährung und Stoffwechsel und erfand einen Respirationsapparat zur Messung der Atemgase, den 'Tigerstedt-Sonden Respiratonsapparat'. Zusammen mit seinem Studenten Per Bergman entdeckte er 1898 das Enzym 'Renin'; seine Lehrbücher wurden Standardwerke. Ab 1890 ging er für zwei Jahre nach Strassburg zu Schmiedeberg und nach Leipzig zu Böhm. Nach seiner Rückkehr lehrte er Pharmakologie in Uppsala, 1895 wurde er zum a.o.
