Methotrexat ist ein Krebsmedikament. Es hat aufgrund seiner schnellen…Freisetzung im Magen-Darm-Trakt (GIT) unerwünschte Nebenwirkungen. Das schwimmende In-situ-Gel wurde unter Verwendung von In-situ-gelbildenden Polymeren wie Natriumalginat und Pektin hergestellt. Die Kompatibilität von Wirkstoff und Hilfsstoffen wurde mittels FT-IR-Untersuchung geprüft. Zur Untersuchung der kombinierten Wirkung der Natriumalginatkonzentration (X1) und der Calciumcarbonatkonzentration (X2) wurde ein 3-stufiges 2-Faktoren-Vollfaktorielles Design angewendet. Die schwimmenden In-situ-Gele wurden hinsichtlich ihrer Schwimmverzögerungszeit, der prozentualen Wirkstofffreisetzung nach 1 Stunde und der prozentualen Wirkstofffreisetzung nach 12 Stunden bewertet. Zusätzlich zu den oben genannten Parametern wurden die schwimmenden In-situ-Gele auch hinsichtlich pH-Wert, Viskosität, Gelierfähigkeit, Schwimmzeit und Wirkstoffgehalt bewertet. Alle Formulierungen lieferten zufriedenstellende Ergebnisse hinsichtlich der Schwimmverzögerungszeit, der prozentualen Wirkstofffreisetzung nach 1 Stunde und nach 12 Stunden. Die Wirkstofffreisetzung hing von der Konzentration des in der Formulierung verwendeten Polymers und Calciumcarbonats ab.

Methotrexate is anticancer drug. It shows th…e undesirable side effect due to their fast release in the gastrointestinal tract (GIT). Floating in situ gel was prepared by using in-situ gel forming polymers like sodium alginate and pectin. Drug-excipients compatibility was carried out by FT-IR study. A 3level 2factor full factorial design was applied to investigate the combined effect of sodium alginate concentration (X1) and calcium carbonate concentration (X2). Floating In situ gels were evaluated for floating lag time, % drug release at 1 hr and % drug release at 12 hrs. In addition to the above, floating in situ gels were also evaluated for, pH, Viscosity, gelling capacity, floating time, drug content. All formulation gave the satisfactory result in terms of floating lag time, % drug release at 1 hr, and 12 hrs. Drug release was depended on the concentration of the polymer and calcium carbonate used in formulation. 92 pp. Englisch.
