Er befasste er sich insbesondere mit der Physiologie des Blutkreislaufs, Ernährung und Stoffwechsel und erfand einen Respirationsapparat zur Messung der Atemgase, den 'Tigerstedt-Sonden Respiratonsapparat'. Zusammen mit seinem Studenten Per Bergman entdeckte er 1898 das Enzym 'Renin'; seine Lehrbücher wurden Standardwerke.
