This oncogene influences various functions of the AML cell and leads to uncontrolled pro…liferation. Clinically, FLT3-ITD is associated with a poor prognosis in patients with AML. In this study, the influence of FLT3-ITD on the phosphatase CDC25C, which as a cell cycle regulator significantly influences the control of cell division, was investigated. The experiments described here showed that CDC25C is phosphorylated by FLT3-ITD and activated in an uncontrolled manner. In the leukemia cell model, a primarily protective effect of CDC25C was shown in the presence of oncogenic stress. The genetically non-intact cells were increasingly prevented from dividing by CDC25C. However, the simultaneous presence of the FLT3-ITD mutation resulted in a loss of this protective function. The significance of the mechanism described here therefore lies in the control of the response of the leukemia cell to damage within the DNA. Los experimentos aquí descritos mostraron que CDC25C es fosforilada por FLT3-ITD y activada de forma incontrolada. En el modelo celular de leucemia, se demostró un efecto principalmente protector de CDC25C en presencia de estrés oncogénico. CDC25C impidió cada vez más la división de las células genéticamente no intactas. Sin embargo, la presencia simultánea de la mutación FLT3-ITD provocó una pérdida de esta función protectora. Cet oncogène influence différentes fonctions de la cellule LAM et entraîne…une prolifération incontrôlée. Cliniquement, la FLT3-ITD est associée à un mauvais pronostic chez les patients atteints de LAM. Dans ce travail, l'influence de la FLT3-ITD sur la phosphatase CDC25C, qui, en tant que régulateur du cycle cellulaire, influence de manière déterminante le contrôle de la division cellulaire, a été étudiée. Les expériences décrites ici ont pu montrer que CDC25C est phosphorylée et activée de manière incontrôlée par la FLT3-ITD. Dans le modèle de cellules de leucémie, un effet protecteur primaire de CDC25C a été observé en présence d'un stress oncogène. Les cellules génétiquement non intactes ont été davantage empêchées de se diviser par CDC25C. Cependant, la présence simultanée de la mutation FLT3-ITD a entraîné une perte de cette fonction protectrice. L'importance du mécanisme décrit ici réside donc dans le contrôle de la réponse de la cellule leucémique aux dommages au sein de l'ADN.
