El aumento de la solubilidad se obtuvo a travé…s de su complejación con CD. Los sistemas de DH-M¿CD permitieron una notable mejora de la solubilidad inicial del fármaco en el agua. En particular, se obtuvo un aumento de unas 22 veces para el producto amasado. Los estudios de DSC, FTIR y PXRD indicaron que los complejos pueden formarse por el método de amasado. La utilización de esos sistemas binarios de drogas-CD preparados por el método de amasado permitió la preparación de tabletas bucales directamente comprimibles que mostraron mejores estudios de células de difusión y valores de disolución en comparación con la formulación comercial y las tabletas que contenían la droga sola. Así pues, puede concluirse que el método de amasado puede utilizarse para preparar complejos DH-M¿CD, a fin de mejorar el perfil de solubilidad y disolución de la droga, que puede mejorarse aún más convirtiendo los complejos en una formulación bucal.

Assim, pode-se concluir que o método de amassamento pode ser usado para preparar complexos DH-M¿CD, para melhorar o perfil de solubilidade e dissolução do fármaco, que pode ser ainda mais aprimorado com a conversão dos complexos em uma formulação bucal.

Enhancement of solubility was obt…ained through its complexation with CDs. DH-MbetaCD systems allowed a marked improvement of the initial drug water solubility. In particular, an increase of about 22 times was obtained for kneaded product. DSC, FTIR, and PXRD studies indicated that complexes can be formed by kneading method. The use of such drug-CD binary systems prepared by kneading method enabled preparation of directly compressible buccal tablets which showed better Diffusion cell studies and dissolution values compared to marketed formulation and tablets containing drug alone. Thus, it can be concluded that the kneading method can be used to prepare DH-MbetaCD complexes, to enhance the solubility and dissolution profile of drug which can be further enhanced by converting the complexes into a buccal formulation. Receptors on the floor of the ventricle of the brain represent a chemo-receptor trigger zone, known as the a…rea postrema, stimulation of which can lead to vomiting. Chlorpromazine HCl is formulated as a sublingual film. The films formed by HPMC K-15 at a concentration of 120 mg showed the desirable character for a sublingual film. The optimized films formed were subjected to evaluation parameters viz. Tensile Strength, Disintegration, In-vitro dissolution, Ex-vivo Permeation studies and In-vivo studies and the results complied with the available statistical data. In-vitro dissolution studies were analyzed and Higuchi model was found to be the best fit model for In-vitro dissolution studies with a regression value of 0.9248. Ex-vivo permeation studies showed fivefold enhanced permeation of drug from films as compared to pure drug. In conclusion, a sublingual film of Chlorpromazine HCl may result in better patient compliance, quick onset of action and enhanced bio-availability. 116 pp. Englisch.
