Many researchers have worked on extraction and characterization of this enzyme from different plants and still working on its inhibition to prevent the loss.In this project, we have made an attempt to extract the enzyme polyphenol oxidase from the pulp of bread fruit and characterise it.The study has revealed the presence of active Polyphenol oxidase in the pulp of A. altilis which has high specificity for catechol compared to phenol, whose activity lost by TCA, which has optimum pH of 6,optimum temperature of 35 degree celcius,the SDS-PAGE and Zymography technique was employed to determine the molecular weight of enzyme. El estudio se llevó a cabo para aislar y caracterizar la polifenoloxidasa del fruto de Artocarpus altilis (fruta del pan). Estas enzimas se encuentran normalmente en los cloroplastos de las plantas, aunque se liberan de este compartimento durante la senescencia o la maduración. Esta enzima es responsable del oscurecimiento de muchas frutas y verduras, lo que supone una pérdida económica para los agricultores. Muchos investigadores han trabajado en la extracción y caracterización de esta enzima de diferentes plantas y siguen trabajando en su inhibición para evitar las pérdidas. En este proyecto, hemos intentado extraer la enzima polifenol oxidasa de la pulpa del fruto del árbol del pan y caracterizarla. El estudio ha revelado la presencia de polifenol oxidasa activa en la pulpa de A. altilis, que tiene una alta especificidad para el catecol en comparación con el fenol, cuya actividad se pierde por el TCA, que tiene un pH óptimo de 6 y una temperatura óptima de 35 grados centígrados. Se emplearon las técnicas SDS-PAGE y zimografía para determinar el peso molecular de la enzima. Lo anterior ofrece una breve perspectiva para futuras investigaciones.…

Beaucoup de chercheurs ont travaillé sur l'extraction et la caractérisation de cette enzyme à partir de différentes plantes et continuent de travailler sur son inhibition afin de prévenir les pertes. Dans le cadre de ce projet, nous avons tenté d'extraire l'enzyme polyphénol oxydase de la pulpe du fruit à pain et de la caractériser. L'étude a révélé la présence de polyphénol oxydase active dans la pulpe d'A. altilis, qui présente une spécificité élevée pour le catéchol par rapport au phénol, dont l'activité est perdue par le TCA, qui a un pH optimal de 6 et une température optimale de 35 degrés Celsius.
