Although various chemotherapeutic agents have been developed, still there is no successful treatment against multidrug resistance acquired…by the cancer cells due to high P-gp expression in the cell walls. In the effort to synthesize new compounds that overcome multidrug resistance, we have synthesized a novel series of N-unsubstituted and N-substituted [1,4]-DHPs and evaluated these derivatives for P-gp inhibitory activity using Rhodamine B as P-gp substrate. From the in vitro assay, it has been found that N-substituted [1,4]-DHPs exhibits improved activity then N-unsubstituted compounds. The aliphatic substitution at 1st position of unsymmetrical [1,4]-DHPs resulted in good fluorescence uptake.

En un esfuerzo por sintetizar nuevos compuestos que superen la resistencia a múltiples fármacos, hemos sintetizado una nueva serie de [1,4]-DHP sin sustituir en N y sustituidos en N, y hemos evaluado la actividad inhibidora de la P-gp de estos derivados utilizando rodamina B como sustrato de la P-gp. A partir del ensayo in vitro, se ha observado que los [1,4]-DHP sustituidos en N muestran una actividad mejorada en comparación con los compuestos no sustituidos en N. La sustitución alifática en la primera posición de los [1,4]-DHP asimétricos dio lugar a una buena captación de fluorescencia.

Dans le but de synthétiser de nouveaux composés capables de surmonter la multirésistance aux médicaments, nous avons synthétisé une nouvelle série de [1,4]-DHP non substituées en N et substituées en N, et évalué l'activité inhibitrice de la P-gp de ces dérivés en utilisant la rhodamine B comme substrat de la P-gp. Les essais in vitro ont montré que les [1,4]-DHP N-substitués présentent une activité supérieure à celle des composés N-non substitués. La substitution aliphatique en position 1 des [1,4]-DHP asymétriques a permis d'obtenir une bonne absorption de la fluorescence.
