Elle les testa d'abord sur les malades des hopitaux avant de les servir à son père, qui désapprouvait sa liaison. Celui-ci liquidé, elle se débarrassa de ses deux frères, de sa sur carmélite. Sainte-Croix mourut en 1672, laissant les recettes de la marquise, sa confession et des fioles de poison. Condamnée à mort, elle eut une fin exemplaire et passa même pour sainte. Mme de Sévigné évoque l'affaire à de nombreuses reprises dans ses lettres : « On ne parle ici que des discours et des faits et gestes de la Brinvilliers. A-t-on jamais vu craindre d'oublier dans sa confession d'avoir tué son père ? Les peccadilles qu'elle craint d'oublier sont admirables. Elle aimait ce Sainte-Croix, elle voulait l'épouser, et empoisonnait fort souvent son mari à cette intention. Sainte-Croix, qui ne voulait point d'une femme aussi méchante que lui, donnait du contre-poison à ce pauvre mari ; de sorte qu'ayant été ballotté cinq ou six fois de cette sorte, tantôt empoisonné, tantôt désempoisonné, il est demeuré en vie et s'offre de venir solliciter pour sa chère moitié. » (à Mme de Grignan, 1er mai 1676).
