Druck auf Anfrage Neuware - Printed after ordering - Das Hämoglobin ist eines der wenigen Proteine, bei dem einerseits die Beziehungen zwischen Struktur und Funk tion bis in den molekularen Bereich hinein bekannt sind und andererseits die genabhängige Determination einer jeden Aminosäure im Globintetrameren sowie deren mutative Veränderungen detailliert analysiert wurden. Mit dem Nachweis der Hämoglobinvariante Hb S bei der Sichelzellanämie (PAULING et al. 1949) und ihrer Struk turaufklärung (INGRAM 1957) wurde erstmals der direkte Beweis für die Synthese eines veränderten Proteins infolge einer Genmutation erbracht. Durch die Sequentierung der entsprechenden mRNA ist schließlich im Jahre 1977 (MAROTTA et al. 1977) die zugrunde liegendemutative Ver änderung im Gen aufgeklärt worden. So haben protein chemische, molekularbiologische und physiologische Un tersuchungen am normalen Hämoglobin und an seinen Varianten zur Lokalisierung der Basisdefekte der Hämo globinopathien bis hin zu den NUKleotidveränderungen im mutierten Gen geführt. Bisher sind mehr als 300 Varianten des Hämoglobins beschrieben und ihre Molekularpathologie untersucht worden. PAULING prägte für die durch diese Varianten be dingten Hämoglobinopathien den Begriff der Molekular krankheit. Dem raschen Fortschritt der letzten beiden Jahre auf dem Gebiet der Molekulargenetik der Hämoglobinsynthese Rechnung tragend, soll in vorliegender kurzer Mono graphie eine geschlossene Darstellung der Struktur Funktions-Beziehungen und der Genetik des normalen Hämoglobins des Menschen und seiner Varianten sowie Vorwort 6 eine Pathophysiologie ausgewählter Hämoglobinopathien gegeben werden, die sowohl dem Mediziner als auch dem Naturwissenschaftler die molekularen Aspekte einer Gruppe sehr gut untersuchter genetisch bedingter Defekte des Menschen nahe bringen möchte.…
