Neuware -Oxidative stress and aging Over the past several years there has been an extraordinarily rapid growth in our knowledge of free radical chemistry and its possible involvement in both normal essential biology and age related disease and dysfunction. Much of this growth in the traditionally separate sciences of chemistry and molecular gerontology occurred independently, with little interaction or communi cation between the scientists working in these two fields. In view of the growing maturity of the two fields and the potential importance of advancing our knowledge in the area of oxidative stress and aging, we perceived a critical need to organize an international conference the 'First Inter national Conference on Oxidative Stress and Aging' in Hawaii in 1994 to bring together the world's leading scientists in the fields of reactive oxygen species and molecular gerontology. The objective of this conference was to provide a unique opportunity for scholars working in these two related and rapidly growing fields to participate in the exchange, integration, and synthesis of new concepts and ideas, to engage in constructive criticism and to initiate new collaborative research projects. The conference focused on the molecular and cellular aspects of aging as related to oxidative stress. It was one of the largest and most comprehensive international conferences held in molecular gerontology. In the 1970s the H2-receptors became important targets for developing new anti-ulcer agents. The H2-antagonist story is a stimulating example of modern research on pharmacology and medicinal chemistry. Unknown and new properties of histamine and histaminergic systems came to the attention of the pharmacologists and medicinal chemists in the 1980s. The role of histamine as a neurotransmitter, the existence of a presynaptic receptor (H ) controlling the release of histamine 3 and other neurotransmitters, and the particular character of this receptor became important as the possibilities to design new therapeutic agents were realized. However, in many countries their popularity is waning as prescribers become more aware of problems with unwanted effects and with rebound and withdrawal syndromes. The earlier benzodiazepines were long-acting, and it was thought that their shorter-acting successors would have important advantages. Other developments include partial agonists and partial inverse agonists, compounds acting on the benzodiazepine-GABA macro molecular site although not as a typical benzodiazepine, and anxiolytic agents acting on 5-HT pathways directly. I shall review the problems of unwanted effects and dependence, mainly from an epidemiological view point, and then relate the pharmacological properties of the newer com pounds to possible differences in their clinical profile of adverse effects. Adverse behavioural effects Benzodiazepines are used, inter alia, as anxiolytics and hyp notics. The latter use exploits the sedative actions which occur at higher than anxiolytic doses. The sedation should disappear by the time the patient wakes up. In the last two decades, some extensions of the finite difference method to irregular networks have been described and applied to solving boundary value problems in science and engineering. For instance, 'box integration methods' have been widely used in electro nics. There are several papers on this topic, but a comprehensive study of these methods does not seem to have been attempted. The purpose of this book is to provide a systematic treatment of a generalized finite difference method on irregular networks for solving numerically elliptic boundary value problems. Thus, several disadvan tages of the classical finite difference method can be removed, irregular networks of triangles known from the finite element method can be applied, and advantageous properties of the finite difference approxima tions will be obtained.
