What if the brain has a built-in mechanism that keeps track of how long we have been awake—and eventually makes sleep impossible to resist?
For decades, neuroscientists have known that prolonged wakefulness increases the biological drive to sleep. After sleep deprivation, we do not simply sleep longer. We often sleep more deeply, as if the brain were compensating for something that has accumulated during wakefulness.
But one fundamental question remained unanswered:
Where does that pressure come from?
On August 19, 2026, researchers at the Biozentrum of the University of Basel published a groundbreaking study in Nature that brought science significantly closer to an answer.
Led by William Joo and Alexander F. Schier, the Basel team identified neuronal populations that become activated during prolonged wakefulness and play a crucial role in regulating the drive to sleep. Using whole-brain activity mapping, genetically targeted manipulation and electrophysiology in mice, the researchers identified specific populations in the anterior medial preoptic area and median raphe that respond to prolonged wakefulness and influence subsequent sleep.
When specific neurons were activated, the animals experienced longer and deeper recovery sleep. When particular GABAergic and serotonergic neurons were inhibited, sleep was dramatically reduced—by nearly 70% in the long-term manipulation.
The discovery does not mean that humans can safely sleep only a few hours a night. It is not a treatment for insomnia, nor does it mean that scientists have “solved” sleep.
It reveals something more fundamental: a previously elusive part of the neural machinery that helps generate the sleep drive.

To understand why the Basel discovery matters, this book travels through the history of modern sleep neuroscience: Borbély's two-process model, circadian regulation, homeostatic sleep pressure, the neural circuits of wakefulness and sleep, the flip-flop model, orexin, adenosine, sleep deprivation and recovery sleep.
Then the story enters the laboratory.
How can researchers search the brain for neurons that respond specifically to prolonged wakefulness? How can they distinguish sleep-drive mechanisms from ordinary wakefulness, stress or recovery? And what happens when those neurons are switched on or off?
These questions lie at the heart of the 2026 discovery.
BEYOND THE HEADLINEThe significance of this research extends beyond the headlines.
Understanding the neural mechanisms that generate sleep drive could eventually open new avenues for studying insomnia, sleep disorders, chronic sleep deprivation and the brain's ability to adapt to prolonged wakefulness. But those possibilities remain future questions, not established treatments.
That distinction matters.

Statutory right to withdraw
You have the right to withdraw from this contract within 14 days without giving any reason.
The withdrawal period will expire after 14 days from the day on which you acquire, or a third party other than the carrier and indicated by you acquires, physical possession of the last good or the last lot or piece.
To exercise the right of withdrawal, electronically fill in and submit a clear statement on our website, under "My Purchases" in "My Account". We will communicate to you an acknowledgement of receipt of such a withdrawal on a durable medium (e.g. by e-mail) without delay.
To meet the withdrawal deadline, it is sufficient for you to send your communication concerning your exercise of the right of withdrawal before the withdrawal period has expired.
Effects of withdrawal
If you withdraw from this contract, we will reimburse to you all payments received from you, including the costs of delivery (except for the supplementary costs arising if you chose a type of delivery other than the least expensive type of standard delivery offered by us).
We may make a deduction from the reimbursement for loss in value of any goods supplied, if the loss is the result of unnecessary handling by you.
We will make the reimbursement without undue delay, and not later than 14 days after the day on which we are informed about your decision to withdraw from this contract.
We will make the reimbursement using the same means of payment as you used for the initial transaction, unless you have expressly agreed otherwise; in any event, you will not incur any fees as a result of such reimbursement.
