Streptokinase (SK) is a monomeric, multidomain protein of 47 kDa, produced by several beta-hemolytic Streptococci. It has potential therapeutic activity in blood circulatory conditions especially myocardial infarctions . SK is one of the major blood clot dissolving agents used in many medical treatments. SK gene of S. pyogenes was successfully amplified by hot start PCR product was 1353 bp. It was cloned in pTargeT and pSin vectors and clone containing gene in right orientation were selected after RE analysis, PCR and sequencing. The expression of SK gene using both recombinant plasmids showed good immunofluorescence while control cells did not show fluorescence, similarly immunoperoxidase test showed good expression while controls did not show any expression. The modeling of streptokinase protein using SWISS-MODEL and analysed using Rasmol software to visualize the 3-D structure generated backbone, structure color group, ball & stick, space fill, shapely, strand, wireframe and ribbon model structure. The book is specially useful to students of Biotechnology, Microbiology, Researchers, Teachers and Medical Personnel.
