Designing of a successful enzyme responsive DDS responsive DDS requires exquisite bioengineering of the carrier materials so that they are able to respond to a particular enzyme present in the target site. This powerful concept has applied to various DDS for more effective site specific delivery of cargo. Different smart strategies have been adopted to trigger release in response to enzyme catalysis. Incorporation of peptide sequence onto peptide precursor for responsive release by known substrate can be mentioned as instance. Enzyme triggered hydrogel swelling or collapse is another intelligent strategy has depicted here. Among nanoparticles, silica nanoparticles with pororous reservoirs have drawn much attention after the invention of gatekeping concept. In this chapter several exciting DDS such as polymeric assembly, MSNs, QDs, hydrogels, metal nanoparticles have been introduced with selective scientific references. Several smart approaches for each type of DDS in the context of enzyme sensitiveness have been highlighted for better understanding.
