Transfection via Nucleofection is one of the most efficient non- viral method of introducing a foreign gene in to the host cell. Cell-type specific combinations of electrical current and solutions make this technology unique in its ability to transfer polyanionic macromolecules directly into the nucleus. Therefore, even cells with limited potential to divide are made accessible for efficient gene transfer. This monograph shows various optimization parameters and protocols for standardizing nucleofection protocol in goat fibroblast cells using bovine IFN- τ tagged with EGFP as the model gene construct. This technique can be utilized effectively for the production of animals with better production performances. In addition to the brief protocols of various biotechniques, this book offers a general review of the literatures dealing with the different ways of transfection ,assay of transfection, with the hope that it will give compact information to the researchers and students involved in the reproductive biotechnology.
