Drought responsive genes or transcription factors are important components of any genetic engineering or breeding approach to enhance drought tolerance trait of a crop.Water-deficit stress was imposed on 30 days old seedlings of soybean cultivars Hardee and NRC7.Drought associated physiological parameter like relative water content revealed that water status of the stressed plants is low. Similarly studies on membrane damage disclosed plants under moisture-deficit stress exhibited higher electrolytic leakages, hence greater membrane damage.Total RNA isolated from both the control and stressed plant leaves of cultivars Hardee and NRC-7 was subjected to cDNA synthesis using 3 different anchored primers and thus mRNA population is sub-divided in to 3 different cDNA populations.DDRTPCR using the same anchored primers in combination with eight different arbitrary primers exhibited the transcript profiling of cultivars under moisture deficit stress.The transcript profiling was resolved in denaturing sequencing gel PAGE followed by silver staining so as to identify the differentially expressed transcripts.The book sheds light on drought responsive molecular mechanism of soybean.
