Synopsis

Understanding the mechanisms and control of gene transcription is one of the central goals for a large proportion of molecular biologists currently involved in research. This book concentrates on R.N.A. polymerase II transcription of eukaryotic protein-coding systems but many of the approaches and techniques described apply equally well to the study of systems involving R.N.A. polymerases I and III. This book covers all the major current procedures and approaches in this field, including not only the analysis of transcription in vitro and in vivo but also the identification, purification and characterization of transcription factors and their interaction with specific D.N.A. target sites. Appendices list useful reference data including details of all known transcriptional control cis-elements and trans-factors for R.N.A. polymerase II genes, complete with a full bibliography.

Review

'The book should be of great value to both the novice and experienced researcher, and presents a broad variety of methods in one convenient volume. The format of step-by-step protocols makes for easy use, introductory and background comments are included, and the tips and trouble-shooting guides should help the worker obtain meaningful results.'Mark Nichols, Gene Expression Programme, EMBI, Heidelberg, Trends in Genetics, January 1994, Volume 10

'gives practical details how to deal with: assay of gene transcription in vitro' International Journal of Biochemistry, Vol. 25, No. 12, 1993

'if you have the 'will' to study transcription factors this book provides the 'way''Dr G. Bilbe, Cell Biology International, Vol. 17, No. 12, 1993

all the protocols are uniform in their format and are well organized ... Each chapter contains useful general introductory material on what can be achieved by the described protocols ... Chapter 1, entitled "Assay of gene transcription in vitro" is especially well done ... All of the chapters contain extensive and timely references. This book is a useful compilation of methodologies for studying mechanisms of transcriptional regulation. (Nisson Schechter, State University of New York, The Quarterly Review of Biology, Volume 69)
