One is Euler's solution of the Konigsberg Bridges Problem, dated 1736, and the other is the appearance of Denes Konig's textbook in 1936. 'From Konigsberg to Konig's book' sings the poetess, 'So runs the graphic tale . . . ' [10]. There were earlier books that took note of graph theory. Veb len's Analysis Situs, published in 1931, is about general combinato rial topology. But its first two chapters, on 'Linear graphs' and 'Two-Dimensional Complexes', are almost exclusively concerned with the territory still explored by graph theorists. Rouse Ball's Mathematical Recreations and Essays told, usually without proofs, of the major graph-theoretical advances ofthe nineteenth century, of the Five Colour Theorem, of Petersen's Theorem on I-factors, and of Cayley's enumerations of trees. It was Rouse Ball's book that kindled my own graph-theoretical enthusiasm. The graph-theoretical papers of Hassler Whitney, published in 1931-1933, would have made an excellent textbook in English had they been collected and published as such. But the honour of presenting Graph Theory to the mathe matical world as a subject in its own right, with its own textbook, belongs to Denes Konig. Low was the prestige of Graph Theory in the Dirty Thirties. Isoform-specific interactions of apoE therefore become critical in the mechanism of AD pathogenesis. Detailed characterization of the binding of the apoE isoforms with proteins and peptides relevant to the pathology of the disease may be critical in understanding disease pathogenesis. These critical isoform-specific interactions of apoE may involve interactions with proteins and pep tides in the defining neuropathologic lesions of the disease, the neurofibrillary tangle and senile plaque. Other possible critical isoform-specific interactions include the mechanism of internalization, intracellular trafficking, and subsequent metabolism. In addition, differential post-translational modifications of apoE isoforms may determine differences in metabolism contributing to the pathogenesis of the disease. Oxidation of apoE may confer several isoform-specific, biochemically distinct properties. Since {3A peptide binds apoE in the lipoprotein binding domain of the protein and not in the receptor-binding domain, apoE could target bound {3A4 peptide to neurons via the LRP receptor. Internalization of the apoEI {3A peptide complex into the cell, by the same route as the apoE-containing lipoproteins, would result in incorporation into primary lysosomes and pH dependent dissociation. The first meeting took place in Seefeld, Austria, in February 1992; the topics for the first IWCN were chosen to provide a broad foundation of clinical science, which included the problem of aging, classification of neurodegenerative disorders and of Alzheimer's dis natural history, pathology, and clinical neurology ease, Parkinson's disease, and amyotrophic lateral sclerosis. The fundamental pathology underlying these neurodegenerative disorders is neuronal cell death. For the understanding of pathophysiol ogy and the development of neuroprotective treatment for these dis orders, elucidation of the mechanism of neuronal cell death at the cellular and molecular level is essential. With this concept in mind, the second IWCN was held in Whistler Village in Canada in January 1993. As a mathematical physi cist I lack the depth of training in philological-historical disciplines necessary to discuss the sources of Car dano's knowledge and trace the influences that shaped his views on science, medicine, and philosophy. What little recent literature on Cardano there is some times shows a lack of true understanding, or is primarily an appreciation of the mathematician. I relied, therefore, largely on his own writings, which are collected in the Opera Omnia. My excerpts and translations are taken di rectly from these works, and I hope that I have succeeded in capturing their essential meaning and spirit. These models are derived from experimental data, hence they describe real phenomena only approximately. Thus a mathematical approach must begin with choosing properties which are not very sensitive to small changes in the model, and so may be viewed as properties of the real process. By this reason different notions of stability played an important role in the qualitative theory of ordinary differential equations commonly known nowdays as the theory of dynamical systems. In this book we are concerned mainly with questions of global stability in the presence of noise which can be described as recovering parameters of dynamical systems from the study of their random perturbations.
